Description

StemRelease™ is a gentle, enzyme-free cell dissociation reagent designed for the selective detachment and routine passaging of human pluripotent stem cells (hPSCs), including human induced pluripotent stem cells (iPSCs) and embryonic stem cells (ESCs).

This ready-to-use reagent enables rapid colony release while preserving multicellular aggregate integrity, cell viability, and pluripotency. StemRelease™ supports reproducible feeder-free stem cell culture workflows and is optimized for routine maintenance, colony selection, cryopreservation preparation, and downstream differentiation applications.

StemRelease™ is designed to minimize cellular stress associated with enzymatic dissociation and supports stable long-term expansion of undifferentiated hPSC cultures.

StemRelease™ has been validated in feeder-free human pluripotent stem cell workflows using:

  • Vitronectin-coated culture systems
  • Matrix-based coating systems including Matrigel and 3DCelMatrix™
  • Defined and protein-containing stem cell maintenance media

Usage

Recommended Companion Products

Recommended Workflow

  1. Wash cells once with1X DPBS, No Ca²⁺, No Mg²⁺ (CH110).
  2. Add StemRelease™ to culture vessels and incubate briefly.
  3. Aspirate reagent and incubate the plate until colony edges begin to loosen.
  4. Add maintenance medium and gently detach colonies as small multicellular aggregates.
  5. Passage cells onto freshly coated culture vessels.

For optimal recovery, use ROCK Inhibitor Y-27632 (TM131) during stressful handling conditions such as thawing or single-cell applications.
