Cell culture coating solutions are protein or polymer preparations applied to culture surfaces to improve cell attachment, spreading, and viability. Many primary cells, stem cells, and specialized cell types fail to adhere or function properly on uncoated plastic. Coatings provide integrin-binding sites that recapitulate the extracellular matrix environment found in vivo, resulting in better experimental reproducibility and more physiologically relevant outcomes.
What is the difference between 3DCelMatrix™ and Applied Cell Extracellular Matrix?
3DCelMatrix™ (TM076) is a temperature-sensitive basement membrane hydrogel optimized for 3D culture applications. It can be used as an embedded matrix or as a thin surface coating, and is the preferred choice for organoid formation, tumour spheroid culture, and invasion assays. Applied Cell Extracellular Matrix (G422) is a ready-to-use multi-protein ECM coating primarily used for 2D surface coating of demanding adherent cell types, including primary cells and sensitive epithelial lines.
Which coating is best for iPSC and human embryonic stem cell culture?
Vitronectin Coating Solution (TM060) is the recommended choice for feeder-free hESC and iPSC maintenance. Vitronectin binds αVβ5 and αVβ3 integrins that support pluripotency signalling. For iPSC-derived differentiated cell types, such as cardiomyocytes, neurons, and endothelial cells, 3DCelMatrix™ or Applied Cell ECM may provide better results depending on the target lineage.
How do I apply a coating solution to my culture vessels?
Most coating solutions are applied by adding a diluted protein solution to the culture vessel, incubating at room temperature or 37°C for 30–60 minutes, aspirating the excess, and allowing the surface to dry briefly before seeding cells. 3DCelMatrix™ requires cold-chain handling and transitions to a gel at 37°C. Always refer to the product-specific protocol for recommended dilutions and incubation conditions.
Are abm’s coating solutions animal-free or xeno-free?
Synthetic polymer coatings, including Poly-L-Lysine and Poly-L-Ornithine, are animal-component free. Products derived from natural ECM sources, including 3DCelMatrix™ and Applied Cell ECM, contain animal-derived proteins.
