| Description |
High Titer (109 IU/ml) Recombinant Lentivirus co-expressing the R24C mutant of CDK4 along with CCND1 and BMI1 genes from the CAG promoter. The construct also contains a Puromycin cassette for easy cell selection.
Mechanism: One of the main barriers to immortalization (in addition to telomere length) is the p16^INK4a/Rb senescence pathway. This virus contains three components that target this pathway directly — CDK4(R24C) and cyclin D1 keep Rb inactivated, and BMI1 represses the Cdkn2a (p16/p19^ARF) locus — giving a more robust bypass than CDK4(R24C) alone. It is recommended to pair this virus (Cat. No. LV681) with our Lenti-EF1a-hTERT virus (Cat. No. LV616) for maximum immortalization efficiency. |
